Prokaryotic and eukaryotic replicons

چکیده

The replicon model, proposed by Jacob and Brenner (1963) to explain the regulation of prokaryotic DNA replication, has proved remarkably robust. Despite the manyvariations in initiation mechanism discovered since 1963, the replicon model may be applicable, with minor modifications, to regulation of DNA replication in all organisms. For several years, the Replicon Club of Paris, a group of French scientists interested in regulation of replication, has been meeting to discuss new results. This year, with support from the French Centre National de la Recherche Scientifique (CNRS), the Replicon Club sponsored an international Jacques Monod Conference on Prokaryotic and Eukaryotic Replicons. The meeting was organized by G. Buttin (Institut Pasteur, Paris) with assistance from A. Falaschi (International Centre for Genetic Engineering and Biotechnology, Trieste, Italy) and M. Kohiyama (Institut Jacques Monod, Paris) and was held at the CNRS’s Centre Paul Langevin in the Alpine village of Aussois, France, from June 18-22, 1995. There, invigorated by beautiful surroundings and excellent cuisine, scientists from around the world provided evidence both for the vitality of the replicon model and for substantial advances in our understanding of the regulation of replication.

Bidirectional replication from an internal ori site of the linear N15 plasmid prophage.

The prophage of coliphage N15 is not integrated into the chromosome but exists as a linear plasmid molecule with covalently closed hairpin ends (telomeres). Upon infection the injected phage DNA circularizes via its cohesive ends. Then, a phage-encoded enzyme, protelomerase, cuts the circle and forms the hairpin telomeres. N15 protelomerase acts as a telomere-resolving enzyme during prophage DN...

Specific mutations within the AT-rich region of a plasmid replication origin affect either origin opening or helicase loading.

Prokaryotic and eukaryotic replicons possess a distinctive region containing a higher than average number of adenine and thymine residues (the AT-rich region) where, during the process of replication initiation, the initial destabilization (opening) of the double helix takes place. In many prokaryotic origins, this region consists of repeated 13-mer motifs whose function has not yet been specif...

Capsid protein of eastern equine encephalitis virus inhibits host cell gene expression.

Eastern equine encephalitis virus (EEEV) causes sporadic but often severe cases of human and equine neurological disease in North America. To determine how EEEV may evade innate immune responses, we screened individual EEEV proteins for the ability to rescue the growth of a Newcastle disease virus expressing green fluorescent protein (NDV-GFP) from the antiviral effects of interferon (IFN). Onl...

Examination of Prokaryotic Multipartite Genome Evolution through Experimental Genome Reduction

Many bacteria carry two or more chromosome-like replicons. This occurs in pathogens such as Vibrio cholerea and Brucella abortis as well as in many N2-fixing plant symbionts including all isolates of the alfalfa root-nodule bacteria Sinorhizobium meliloti. Understanding the evolution and role of this multipartite genome organization will provide significant insight into these important organism...

Different mechanistic requirements for prokaryotic and eukaryotic chaperonins: a lattice study

MOTIVATION The folding of many proteins in vivo and in vitro is assisted by molecular chaperones. A well-characterized molecular chaperone system is the chaperonin GroEL/GroES from Escherichia coli which has a homolog found in the eukaryotic cytosol called CCT. All chaperonins have a ring structure with a cavity in which the substrate protein folds. An interesting difference between prokaryotic...
